I just ran into the same thing.
Adding config.action_controller.relative_url_root = "/" to the environment file will at least allow it to start, but most of the links will be broken.
Below is a hack from Collective Idea's Brand Keepers that might work if you add :path_prefix => "/subURI" as an option. I think would you still have to mess with the routes to make it work.
I'll look into it more, but I might just use a sub domain instead.

Right, so the backend tabs don't work because they are coded to /admin/plugin_directory - is there a patch that we can make to that code to support this feature?
