Just start a new project by ng new my-ng-app.

Then add pug pug-loader,
and chang template to template: require('pug-loader!./app.component.pug')()

finally I got a error message here:
ERROR in src/app/app.component.ts(5,13): error TS2304: Cannot find name 'require'.

ng serve --aot produces an error:

ERROR in Error: No template specified for component AppComponent
src/app/app.component.ts(6,14): error TS2304: Cannot find name 'require'.

Additionally, WebStorm’s Angular plugin can’t navigate to such a references and open templates. Hence, usages of the TypeScript methods and properties can’t found in pug files too.

So, frankly speaking, because of these downsides, this is not an option.
Even using vanilla pug-cli will be better choice.
