Normally modules are stored in the "node_modules" directory, so node will be looking for your module there. You can provide an absolute path to your module and it should find it then. deskShell.appDir holds the directory your app.desk file is launched from. So try changing your code to the following:
var mymodule = require( deskShell.appDir + "/mymodule/build/Release/mymodule");
Hopefully it will now find your module.
